ABSTRACT:
Tazarotene is a third-generation topical retinoid used to treat psoriasis and acne vulgaris. When applied topically, this prodrug quickly transforms into its active form, tazarotenic acid. Tazarotene modulates gene expression related to keratinocytedifferentiation, proliferation, and inflammation by selectively binding to retinoic acid receptors (RAR-ß and RAR-?). A common semi-synthetic lincosamide antibiotic for topical acne treatment is clindamycin. It stops bacterial protein production by attaching itself to the 50S ribosomal subunit of vulnerable bacteria. Cutibacterium acnes (formerly known as Propionibacterium acnes), a crucial organism in the pathophysiology of acne, is especially susceptible to Clindamycin's effects. In topical formulations, Tazarotene and Clindamycin work in tandem to provide a complimentary mode of action. To evaluate how well combination therapy and monotherapy reduce both inflammatory and non-inflammatory lesions. To assess the place of antibiotics and topical retinoids in the current acne therapy protocols. For both pharmaceuticals in bulk and topical forms, validation experiments conducted in accordance with ICH Q2(R1) requirements show satisfactory accuracy, precision, linearity, LOD, and LOQ. For simultaneous estimate of high sensitivity, specificity, and reproducibility, RP-HPLC techniques are frequently chosen. Targeting several pathogenic causes of acne, such as inflammation, bacterial growth, and aberrant follicular keratinization, the combination therapy offers synergistic advantages. Clinical results show that topical gels or creams provide better therapeutic results than monotherapy and increase patient compliance.
