ABSTRACT:
Baricitinib is an orally administered, selective Janus kinase (JAK1 and JAK2) inhibitor widely prescribed for the management of rheumatoid arthritis and other inflammatory disorders. Owing to its expanding clinical applications and therapeutic importance, the development of robust, precise, and stability indicating analytical methods is essential to ensure its quality, safety, and efficacy in pharmaceutical formulations. This review comprehensively summarises the analytical approaches reported for the estimation of Baricitinib in bulk drug and dosage forms, with particular emphasis on reverse-phase high-performance liquid chromatography (RP-HPLC). The discussed RP-HPLC methods demonstrate excellent performance characteristics in accordance with ICH validation guidelines, including accuracy, precision, linearity, specificity, range, limit of detection (LOD), limit of quantification (LOQ), robustness, and ruggedness. In addition to chromatographic techniques, complementary methods such as UV–Visible spectroscopy, UHPLC, HPTLC, and LC-MS/MS are also highlighted. Overall, validated analytical methodologies play a critical role in pharmaceutical quality control, regulatory compliance, and stability assessment, ensuring consistent therapeutic performance of Baricitinib-containing formulations.
