ABSTRACT:
Favipiravir, a pyrazine carboxamide derivative with broad- spectrum antiviral activity, has gained significant attention for the treatment of RNA viral infections such as influenza and COVID-19. A precise, accurate and robust Ultra- High-Performance Liquid Chromatography (UHPLC) method was developed and validated for the quantitative estimation of favipiravir in bulk and pharmaceutical dosage forms. Chromatographic separation was performed on a reverse-phase column using an optimized mobile phase under isocratic conditions at a controlled flow rate, with UV detection at a suitable wavelength. The retention time of favipiravir was found to be consistent with good peak symmetry. The method was validated in accordance with ICH Q2(R1) guidelines. Linearity was observed over a suitable concentration range with a correlation coefficient (R²) greater than 0.999. The method demonstrated acceptable accuracy with percentage recovery within 98–102% and precision with %RSD less than 2%. The LOD and LOQ values indicated adequate sensitivity of the method. Robustness studies confirmed the reliability under small deliberate variations in method parameters. The validated method was successfully applied for the assay of favipiravir in marketed tablet formulations, showing no interference from excipients. The developed UHPLC method is simple, rapid, and suitable for routine pharmaceutical analysis.
